Описание: Modulation of presynaptic M4 receptors in the brainstem and postsynaptic M1 receptors in the cortex leads to reduced dopaminergic activity in the nucleus accumbens (reducing positive symptoms), increased dopaminergic activity in the cortex (reducing non-positive symptoms), with no effect on the nigrostriatal pathway (thus no motor effects). With positive clinical trial data continuing to emerge, M4 and M1 receptor agonisists and positive allosteric modulators are on the cusp of potential FDA approval and may bring about a dynamic paradigm shift when it comes to treating all symptom domains of schizophrenia (both positive and non-positive) without the motor side effects that can be debilitating and lead to treatment discontinuation.
