The amyloid-beta (Aβ) protein is cut from a larger protein called the amyloid precursor protein. The leading hypothesis regarding the neurobiological progression of Alzheimer's Disease posits that the Aβ protein begins to accumulate in the brain with age due to increased production or decreased degradation of Aβ and/or as a result of genetic and/environmental factors. As Aβ accumulates, it also causes activation of several kinases; these kinases cause hyperphosphorylation of tau. As AD progresses, synaptic dysfunction spreads. There are several environmental and lifestyle factors that appear to convey some protection against the development of Alzheimer's Disease, such as social engagement and exercise.
