Protocol for Freezing iPSCs

In this video, we are going to go over the steps to freeze induced pluripotent stem cells (iPSCs). Different types of freezing media can be used to freeze these cells. In this protocol, cells are frozen in a final solution of 10% Dimethyl sulfoxide (DMSO) in Fetal Bovine Serum (FBS). Keep in mind that iPSCs thrive as colonies, so it is important to carefully manipulate these cells as to not form a single cell suspension.
List of materials:
Cryotubes
20% dimethylsulfoxide (DMSO) in fetal bovine serum (FBS)
FBS
Cryo-box
-20 °C freezer
-80 °C freezer
Liquid nitrogen tank
Before starting this protocol, follow the steps of one of our passaging protocols up to the centrifugation step. For more information, see the passaging protocol videos. In the previous video, the cell suspension was transferred to a conical centrifuge tube and centrifuged for 3 minutes at 1,200 RPM.
