In this video, we are going to learn another protocol for passaging induced pluripotent stem cells (iPSCs). This time we will demonstrate the passaging of cells with ReLeSR. ReLeSR is an enzyme-free dissociation reagent used for passaging iPSCs. It comes to the rescue when spontaneously differentiated cells exceed 25-30% of the total number of cells, making it difficult to manually remove these areas.
List of Materials:
Phase-contrast microscope
Matrigel-coated dish
ReLeSR
Dulbecco's phosphate-buffered saline (D-PBS)
iPSC Media (mTeSR, mTeSR plus or E8)
5 mL and 10 mL plastic and glass serological pipettes
Sterile pipette tips
ROCK inhibitor
