Описание: This research study presents a large-scale genomic analysis of breast cancer by examining over 9,500 patient samples to identify critical genetic drivers. The authors categorized 1,174 significantly mutated genes into five priority tiers based on how often they occurred across nineteen clinical studies. Their findings highlight PIK3CA and TP53 as the most frequent mutations, occurring in over 30% of cases, followed by other key markers like GATA3 and CDH1. By mapping these genetic alterations to signaling networks, the study identifies the PI3K-AKT, NOTCH, and HIPPO pathways as central to tumor development and drug resistance. Furthermore, the analysis reveals that current commercial testing panels only include a small fraction of these influential genes. Ultimately, this work provides a detailed protein-protein interaction framework to improve personalized diagnosis and therapeutic targeting.
