Certain substituted pyridines can be obtained via rearrangement of pyridine N-oxides (also known as Boekelheide reaction) or via rearrangement of an alternative heterocycle system, ie., pyrrole (is called as Ciamician-Dennstedt reaction).
Boekelheide reaction
This reaction involves the treatment of pyridine N-oxides with acylating agents to form rearranged products. Traditionally, the rearrangement occurs at the a-position, but oxygen migration could also occur on the alkyl chain of substituents attached to the g-position. This process requires the activation of pyridine N-oxides by electrophilic agents such as acetic anhydride (as shown in the reaction), in some cases phosphorous pentoxide (P2O5), trifluoro acetic acid (TFAA), or tosyl chloride (TsCl) are used.
Ciamician-Dennstedt rearrangement
This reaction involves the addition of intermediate dihalocarbene, generated from haloforms (CHX3, X=Cl, Br, or I) and a strong base, to a pyrrole (330) to form an unstable dihalogenocyclopropane, which rearranges to a 3-halogenopyridine such as 3-chloropyridine. The reaction was expanded for indoles to provide 3-chloroquinolines.
