Fullmetal Alchemist: Brotherhood adapts all 27 volumes of the manga, unlike the first television series. Brothers Edward and Alphonse Elric are raised by their mother Trisha Elric in the remote village of Resembool in the country of Amestris. Their father Hohenheim, a noted and very gifted alchemist, abandoned his family while the boys were still young, and while in Trisha's care they began to show an affinity for alchemy. However, when Trisha died of a lingering illness, they were cared for by their best friend Winry Rockbell and her grandmother Pinako.

A Homunculus (人造人間ホムンクルス, Homunkurusu, Latin for "Little Human", Japanese for "Artificial Human") refers to the medieval legendary concept of an artificially created human, presumably brought into existence by certain means of alchemy.

Greed (グリード, Gurīdo) is the embodiment of some of Father's greed. He is called the "Ultimate Shield" because he can rearrange the carbon in his body to make his skin as hard as diamond, thus making him impenetrable by most weapons.

Envy (エンヴィー Envī?) is the embodiment of some of Father's envy. Envy is genderless, and has the ability to shape-shift. This ability can be used to assume the form of any existing person (or original personas that Envy themselves creates). They can even modify their own voice to directly copy others.

Lust (ラスト, Rasuto) is the embodiment of Father's lust. She is mildly flirtatious, but serves mostly as an object of lust for others, feeding their desires so that she could fulfill her duties to Father. She was usually partnered with Gluttony to whom she appears to have a particular (almost motherly) closeness.

Gluttony is the embodiment of some of Father's gluttony. Often, he will be found eating a random person or creature's remains unless ordered to eat someone that poses a threat, or to remove evidence of the Homunculi's presence. He is typically in the company of Lust.

Sloth (スロウス, Surōsu) is the embodiment of some of Father's sloth

King Bradley (キング・ブラッドレイ Kingu Buraddorei?) is one of the primary antagonists in the Fullmetal Alchemist series, being that he is one of the seven Homunculi. Bradley was also the original Führer of Amestris.

Selim Bradley (セリム・ブラッドレイ, Serimu Buraddorei) is the son of King Bradley, the Führer of Amestris. Due to the divergence between the manga and the 2003 anime series, Selim is portrayed as two very different characters in both the continuities. In the manga and 2009 series, he was eventually revealed to be Pride (プライド, Puraido), the first Homunculus that Father created, embodying the cruelest, most prejudicial, yet most basic aspect of evil

The Dwarf in the Flask: Homunculus, referred to as "Father" (お父様, Otō-sama) by his "children", the other homunculi, is the central antagonist of the Fullmetal Alchemist.
