Learn to identify L4s, the 4th larval stage of C. elegans. During this stage, the vulval cells undergo characteristic morphological changes, which makes the vulva appear as a light-colored “half-moon” on one side of the middle of the worm. (Worms actually swim on their sides, so the vulva is technically on the bottom, or “ventral” side of the worm!)
Why pick L4s? This stage is ideal for setting up genetic crosses, as the animals will not have mated with a male prior to the cross (due to the underdeveloped L4 vulva) and will reach egg-laying maturity in the presence of the selected male.
Picking the L4 stage is also a great way to synchronize animals that will become adults by the next day. Many methods will reference this time point (i.e. “Animals were dissected 26 hours post-L4”) so it’s important to be able to properly identify L4 worms!
