In this video, I look at DNA sequencing, starting with the widely used Sanger or chain-terminating dideoxynucleotide method.

If you're familiar with PCR, you'll grasp how this method works as I break down the process into two stages: the reaction and the separation. I look at the role of dideoxynucleotides (ddNTPs) in the reaction, how they terminate DNA chains with fluorescent markers, and how the sequence is ultimately determined.
I also look at the limitations of the Sanger method and how advanced techniques like next-generation sequencing (NGS) and third-generation sequencing are revolutionising DNA analysis, making large-scale projects and rapid sequencing possible.
