The Skraup synthesis is a chemical reaction used to make quinolines, and is named after Czech chemist Zdenko Hans Skraup. The reaction involves heating aniline, glycerol, sulfuric acid, and an oxidizing agent like nitrobenzene. The steps of the reaction are:
Dehydration: Glycerol dehydrates with sulfuric acid to produce acrolein.
Addition: Aniline adds to acrolein to produce compound A.
Ring closure: Compound A undergoes ring closure to form 1,2-dihydroquinoline.
Oxidation: 1,2-dihydroquinoline is oxidized with nitrobenzene to produce quinoline.
The Skraup synthesis is the most common method for making quinolines. A modified Skraup synthesis can be used to make quinolines with substitutions in the hetero ring. This involves using a substituted acrolein or vinyl ketone instead of glycerol.
