Taxonomic assignment: Amoebozoa: Evosea: Cutosea: Squamamoebidae
Cutoseans are tiny amoebae with the cells enclosed in a flexible layer of flat, oval scales embedded in matrix. Four monotypic (i.e. containing a single species only) genera are known: marine Squamamoeba Kudryavtsev and Pawlowski, 2013, Armaparvus Schuler and Brown, 2019, Sapocribrum Lahr et al., 2015, and freshwater Idionectes Hess et al., 2019. By contrast, Armaparvus browni, the second member of Armaparvus, originates from the continental brackish-water habitat – Соляной ручей (“Saline Stream”) oxbow with water flowing from Царицынский источник ("Tsaritsynsky Spring"), a fountain of underground brackish water (19 ppt) on the territory of the town of Staraya Russa (Старая Русса), Novgorod Region, northwest of Russia. Cutoseans were never observed in such biotopes before.

Location: filtered material from water, oxbow of the Solyanoy (“Saline”) Stream in the town of Staraya Russa (Novgorod Region, North-Western Russia; 57.9848N, 31.33846E), salinity ca. 15 ppt (https://www.google.com/maps/d/edit?hl.... Amoeba isolated and cultured in 30 ppt with wheat grains, but tolerates a range of salinities down to 2.5 ppt.
Reference culture: ZIN.2023.15, kept with the Culture Collection of Heterotrophic Protists of the Zoological Institute, Russian Academy of Sciences.
Imaging: Nikon Eclipse Ni upright microscope 100x planapochromate oil immersion object lense with a 1.0x camera adaptor, differential interference contrast.
Observation conditions: Amoebae picked out from the culture, placed on a coverslip in a drop of medium and allowed to settle in a moist chamber. Then the coverslip is placed on a slide with the sealing petroleum jelly border to prevent the preparation from drying out too quickly. The slide is then placed on the microscope with the coverslip up. Observation starts immediately.
