This challenge is a bit tricky. In order to complete it, you must kill an enemy with a Tomahawk by normally throwing it, another one with a Tomahawk melee attack, and another one while you're on horseback.
What I suggest you to do is start a gang hideout or bounty hunter mission. Take out most of the enemy except like maybe 2 or 3 so you don't get overwhelmed and killed. The good thing about bounties is that waves of enemies will keep respawning after you caught/killed the bounty, which is a good thing.
