Daily Growth Promotion Testing (GPT) of microbiological culture media is a critical quality control procedure performed to ensure that each batch of media—whether prepared in-house or purchased pre-poured—is capable of supporting the growth of target microorganisms.
It confirms that the nutrients, sterilization, and storage conditions have not rendered the media, or specific batches of it, inhibitory.
Here is why daily or batch-specific GPT is necessary:
1. Ensuring Microbial Recovery (Sensitivity)
2.Support Growth: The primary purpose of GPT is to verify that the media can support the growth of specific microorganisms, including fastidious (fussy) organisms.
3. Low-Level Challenge: GPT uses a low concentration of microorganisms (typically 10 to 100 Colony Forming Units - CFU) to ensure the medium is sensitive enough to detect low levels of contamination.
4. Percentage Recovery: The media must show adequate growth, often with a recovery range of 50% to 200% compared to a control, to ensure accurate, quantitative results.
