A Danish newspaper claims at least nine of 43 children, fathered by the man's sperm, have been diagnosed with neurofibromatosis type 1.
The disorder causes tumours to grow around nerves.
Nearly 20 of the children live in Sweden and Norway.
Danish health officials say the sperm bank could be shut down for allegedly not disclosing details about the disease when they came to light.
