Gain-staging is dependant on how much signal you'l drive into the summing mixer (generally to taste) to increase or decrease harmonic distortion.
1. Panning - separation: find for all your instruments the right places in to the stereo field, the right locations where they sit better in to the whole mix.
2. Gain staging is like pulling the focus of a camera lens, find for all your instruments/tracks the "gentle distortion" point, where the subtle distortion starts gently and it's about right.
3.Drive the amp - makeup gain - find the sweet-spot when the level and subtle distortion is just about right, depending on the song and production.
