Spirochaete Staining by Negative Staining Method
• Take a clean glass slide.
• Place a small drop of Nigrosine at the end of the slide.
• With the help of toothpick gently scrape out tartar from interspaces between teeth.
• Mix this tooth tartar into the drop of Nigrosine with the help of toothpick.
• Do not spread the drop or let dry.
• Using the end edge of another slide, spread the drop out to produce a smear varying from bluish black to gray.
• Do not heat fix the slide.
• Let the smear air dry.
• Examine under oil immersion lens (100X) of compound microscope.
