In this video I'll show step-by-step how I isolate peripheral blood mononuclear cells (PBMCs) from whole blood using density gradient centrifugation. This should work with Ficoll-Paque™ or Lymphoprep™. I'll also go over common errors or issues, and how you can avoid or fix them.
I'll first go over materials needed, the protocol of layering blood over the density medium to isolate PBMCs, color differences, RBC contamination, and staining issues. Thanks for watching the first video on this channel!
