Staining Prosedure of Digenea and Cestoda

Trematoda/Digenea and Cestoda were fixed in 70% ethanol, stained with acetic carmine (Palm, 2004), dehydrated in an ethanol series, cleared with a graded series of eugenol, and mounted onto slides in Canada balsam.
General Staining Procedure:
1. Fixation: The cestode specimen is fixed to preserve its structure, in alcohol
2. Staining: The fixed specimen is placed in a carmine-based staining solution for an appropriate amount of time, depending on the thickness of the parasite.
3. (Maybe) Clearing/Rinsing: The excess stain is removed, and the specimen is rinsed or cleared with appropriate solutions, such as acid alcohol
4. Dehydration: The specimen was gradually dehydrated using increasing concentrations of alcohol (e.g., 70%, 85%, 90%, and 100%).
5. Mounting: The dehydrated specimen was placed on a clean slide with mounting onto slides in Canada balsam and covered with a coverslip for microscopic examination.
