Two farmers (Royal Dano and John Davis Chandler) discovered the mauled, dead body of a local young woman on Marsh Island in the Louisiana Bayou. Sheriff Aaron Whitaker (David Janssen) is called. Lawrence Burrifors (Geoffrey Lewis), the victim's temperamental brother, arrives at the crime scene and quickly concludes that the girl's lover, whose identity her brother does not know, committed the murder. Dr. Drutan (John Beradino) examines the body and decides that the girl was killed by a blow to the head.
The sheriff is investigating the case, and locals have many views, including the belief that she was killed by wild dogs. A posse forms to track down the wild dogs, but they have little luck. Burrifors continues to insist that the killer is his sister's mysterious boyfriend, but the sheriff grows dubious of him. The sheriff interviews the girl's ailing and dying father, Hugh Burrifors, who warns him about the "Lougarog". The sheriff doesn't comprehend the French phrase.
The sheriff's investigation quickly leads him to the plantation home of the rich Andrew Rodanthe (Bradford Dillman) and his sister Louise (Barbara Rush). They represent the last of a local family dynasty.
Andrew, who the sheriff accuses of having an affair with the victim, claims to have been suffering from malaria the night the girl was slain. The sheriff arrests the volatile brother Lawrence after he strikes the town doctor, who turns out to be the mysterious lover. While there, the full moon rises again, and Lawrence and the sheriff's deputy are killed in a brutal attack when the cell's steel bars are severed from the wall.
With the town's citizens afraid and the sheriff unable to assist, Andrew Rodanthe volunteers to become deputy. Andrew and the sheriff return to Hugh Burrifor's house, where they discover the old man has concocted a voodoo medicine that emits a vapor designed to repel the Lougarog.
Rodanthe inhales the potion and experiences what appears to be an epileptic fit. He is taken to a hospital. While there, Andrew's sister Louise informs the sheriff that she speaks French fluently and would like to speak with Hugh Burrifor about the inexplicable phrase Lougarog. While speaking with the elderly man, Louise solves the conundrum. Lougarog is an incorrect pronunciation of loup-garou. The phrase is translated into English as "werewolf".
Andrew, having been transformed into a werewolf, forcefully flees the hospital and becomes the target of a manhunt. As a posse form, Louise rushes to the mayor, telling him that her brother is ill and that there are medications that can assist. Louise discusses werewolf folklore with Sheriff Whitaker at her and Andrew's home. She exposes a family secret: her grandfather suffered from odd episodes of disease, hinting that he was a werewolf and that Andrew's curse was inherited.
Louise returns to her plantation home alone, and Andrew, still in werewolf form, enters. Louise escapes to the barn. When the werewolf follows, she attempts to set him on fire before returning to the house. Andrew escapes the fire and pursues Louise. She eventually shoots him, just as the sheriff arrives. After his death, Andrew returns to his human form.
