Introduction to Full Spectrum Flow Cytometry - Dr Rui Gardner (Memorial Sloan Kettering Cancer Center). In the last few years, the field of Flow Cytometry has witnessed a technological shift from the classical spectral approach that captures only a fraction of the emitted light to a technology capable of measuring the full spectrum of each fluorochrome present in the cell.
When capturing the full spectrum of a fluorochrome, the additional measured information can be used to better distinguish between dyes with very similar spectral signatures that otherwise would be impossible to pull apart with classical methods. This has led to a significant increase in the number of fluorochromes being used in multiparameter Flow Cytometry panels both in fundamental and clinical research. In addition, Full Spectrum Flow Cytometry also measures the abundance of autofluorescence, which can then be extracted and can lead in certain circumstances to a better signal discrimination between cells expressing and not expressing a certain fluorescent marker.
Yet the democratization of multicolor panel design and background fluorescent extraction come with a price. The added sensitivity of Full Spectrum Flow Cytometry that enables distinguishing very similar dyes, can also detect very small imprecision in staining controls, leading to errors in extracting the correct abundance of each fluorochrome.
To take full advantage of this novel approach, it is therefore important to understand the underlying principles of Full Spectrum Flow Cytometry and how it differs from Classical methods. Adherence to Best Practices in staining, acquisition, and analysis also become critical in applying this technique correctly.
