Many factors affect the transfer of proteins from a gel to a membrane. Follow these recommendations when optimizing transfers of proteins larger than 150 kDa in size.
Gradient gels are ideal for high molecular weight proteins because they leave larger proteins in the more porous part of the matrix, allowing for a more efficient transfer.
Do not to equilibrate an electrophoresed gel in water before transferring.
Because of its higher protein-binding capacity, PVDF usually results in a more efficient transfer than nitrocellulose.
