Active immunotherapy to reduce NOTCH3 accumulation in CADASIL

Helena Karlström, PhD, Karolinska Institutet, Solna, Sweden, shares an overview of her work to develop a novel NOTCH3 immunotherapy for the treatment of CADASIL. In CADASIL, mutations of the NOTCH3 protein lead to its aggregation and subsequent vessel dysfunction and cell degeneration. Her team is focused on reducing these aggregates through active immunotherapy, with initial success in preventing their accumulation in a CADASIL mouse model. Moving forward, the team plans a longer-term therapeutic study to test if active immunization can also reduce aggregates in a more severe mouse model with ongoing pathology, alongside continued research into transcriptomics and proteomics for biomarker discovery. This interview took place at the 10th European Stroke Organisation Conference (ESOC) 2024 in Basel, Switzerland.
