This is the Nathan lab method of generating Differentially Detectable Mycobacterium tuberculosis (DD Mtb) in vitro using two stresses in sequence: starvation in PBS followed by exposure to high dose rifampin. The steps detailed above to generate DD Mtb in this experimental model are demonstrated in a biosafety cabinet without live Mtb cultures. The video is not shot in a biosafety level 3 to facilitate filming, and does not demonstrate all established standard operating procedures normally utilized when working with Mtb.
