FLAURA2 Shows OS Benefit: Dr. Jänne Shares Insights at IASLC World Conference on Lung Cancer

Описание: FLAURA2 Principal Investigator, Pasi A. Jänne, MD, PhD, joined Lung Cancers Today at the International Association for the Study of Lung Cancer (IASLC) 2025 World Conference on Lung Cancer (WCLC) to discuss the final overall survival (OS) analysis from the trial.
David Planchard, MD, PhD, of the Institut Gustave Roussy, presented the findings from the phase 3 trial during the first IASLC Presidential Symposium of the meeting. The phase 3, open-label trial included a total of 557 patients with previously untreated advanced or metastatic non-small cell lung cancer (NSCLC) who harbored exon 19 deletions or L858R mutations in EGFR.
“I’m excited about this morning's presentation of the FLAURA2 final overall survival data,” Dr. Jänne told Lung Cancers Today. “The data was exciting because the combination of chemotherapy and osimertinib led to a median overall survival just shy of four years, which is about a 10-month improvement from that of osimertinib alone. This is a clinically meaningful and significant finding for patients, favoring the combination therapy.”
The median OS was 47.5 months for patients receiving osimertinib plus chemotherapy, compared to the median OS of 37.6 months with osimertinib monotherapy. The 36-month survival rate was 63% in patients receiving osimertinib plus chemotherapy, compared with 51% among those receiving osimertinib monotherapy.
Dr. Jänne explained what these results mean for the treatment landscape in EGFR-mutated NSCLC.
“At the moment, for somebody who gets diagnosed with advanced EGFR-mutant lung cancer, we have monotherapy osimertinib and now two combination therapies, both of which have shown an improvement in progression-free survival and overall survival over single-agent osimertinib,” Dr. Jänne said. “It's nice to have options for patients. We do know however, that the combination therapies have additional side effects compared to single-agent osimetinb.”
This makes it critical to discuss options with patients and engage in shared decision-making, he emphasized.
“For some people, overall survival is the most important endpoint, and it is the gold standard when we think about our effective anti-cancer therapies,” he said. “However… there are people who still prefer single-agent oral therapies.”
Dr. Jänne shared how he counsels patients about combination regimens versus osimertinib monotherapy and explained the key considerations and questions that should be addressed when selecting a therapy.
“When I see patients with newly diagnosed EGFR-mutant lung cancer, we have a discussion about the differences between monotherapy and combinations,” Dr. Jänne explained. “What do the combinations involve? How often does the individual have to come in for infusions and those kinds of things? What are the side effects that come with the added combination and how do we manage those side effects or prevent them from happening in the first place?”
