The first segment details the Seedling Mounting Procedure, illustrating a step-by-step "sandwich" technique. By positioning seedlings between a low-melt agarose blanket and a chambered cover glass, researchers can maintain the root in a stable imaging plane while allowing for natural growth and gas exchange.

The second segment presents the Experimental Result: a 24-hour time-lapse of a regenerating Arabidopsis root following laser ablation. Using three fluorescent reporters (35S:H2B-RFP, WOX5:erCFP, and PET111:YFP), the video captures the dynamic re-establishment of the Quiescent Center (QC) and the reorganization of the WOX5 expression domain in real-time.
