In this webinar hosted by Precision NanoSystems Inc and Cytiva, Dr Ben Peacock describes commonly applied nano-flow cytometry (nFCM) workflows for lipid nanoparticles (LNPs) and shares data from two collaborations evaluating and quantifying RNA payloads.
A - VaxEquity produced self amplifying mRNA (saRNA) LNPs with different ionizable lipids and across a series on N/P ratios (Nitrogen (N) groups in ionizable lipids to phosphate (P) in the RNA).
Particle size, encapsulation efficiency (EE) and PDI remained similar across the N/P ratio but novel nFCM data showed an increase in empty LNPs at higher N/P ratios, even at commonly used N/P ratios.
B - PNI produced mRNA-LNPs across an N/P ratio series and compared formulation processes such as the ignite microfluidic platform and vortex and hand mixing techniques.
Across the N/P ratios, size and EE remains similar and only a population of empty LNPs are observed at higher N/P ratios. However, mRNA quantification showed differences in the RNA payload on a particle by particle basis.
Additionally, when comparing mixing technologies, the microfluidics mixing produced LNPs were highly reproducible, whereas the hand mixed and vortex mixed mRNA-LNPs varied in size and EE batch to batch. nFCM identified that the unencapsulated RNA seen in the vortex and hand mixing samples corresponded with increase populations of empty LNPs, providing additional insight into the lipid-RNA dynamics during LNP formation.
