Molisch Test (General Carbohydrate Test)
Molisch test is a general test for all carbohydrates.
Carbohydrates are dehydrated by concentrated sulfuric acid to form furfural or hydroxymethyl furfural, which reacts with α-naphthol to produce a violet/purple ring at the junction.
Positive for all carbohydrates (mono, di, polysaccharides).
Seliwanoff Test (Ketose Test)
Seliwanoff test is used to distinguish ketoses from aldoses.
Ketoses react faster with resorcinol in HCl to produce a cherry-red color, while aldoses give a slow or faint color.
Positive mainly for fructose and ketose sugars.
Benedict Test (Reducing Sugar Test)
Benedict test detects reducing sugars.
Reducing sugars reduce Cu²⁺ ions to Cu⁺, forming green, yellow, or brick-red precipitate depending on sugar concentration.
Positive for glucose, lactose, maltose.
Quick Exam Trick
Molisch → All carbohydrates
Seliwanoff → Ketose
Barfoed → Mono vs Di
Benedict & Fehling → Reducing sugar.
