Tsunade (綱手) is a Sannin of Konohagakure and a scion of the Senju and Uzumaki Clans. She is renowned as the world's most powerful kunoichi and best medical-nin. Tsunade subsequently abandoned the life of a shinobi for several years due to the recurrent death of her loved ones. She is eventually convinced to return to Konoha and assume the title of Fifth Hokage (五代目火影,, Godaime Hokage, literally: Fifth Fire Shadow), where her abilities prove essential to the community.

Tsunade is Hashirama Senju and Mito Uzumaki's grandchild. Tsunade is referred to as "Princess" (, Hime) since Hashirama was the First Hokage of Konohagakure. Tsunade was Hashirama's first granddaughter, and he found her deviation and adoption of his own gambling addictions immensely funny when she was young. When his father died, Tsunade received his jewellery. Tsunade was partnered with Orochimaru and Jiraiya after graduating from Konoha's Academy, under the supervision of Hiruzen Sarutobi. Tsunade mocked Jiraiya when he failed to obtain a bells during the team's first bell test, causing him to criticise her flat breast.

The beloved Sakura Uchiha (kunohagakure Sakura (née Haruno)) is a notorious kunoichi. After being assigned to Team 7, Sakura quickly discovers she is unprepared for the responsibilities of a shinobi. As a result of training with the Sannin Tsunade, she overcomes this obstacle and is recognized as one of the greatest medical-nin in the world.

Her parents, Mebuki and Kizashi Haruno, have only one child, Sakura. As opposed to her teammates, she had an ordinary childhood, raised by her parents without any serious tragedies or complications. She was picked on by a few girls in her class because of her broad forehead when she entered Konoha's Academy. By hiding her forehead with her bangs, Sakura hoped to combat their teasing, but she proved to them that it bothered her and caused them to tease her even more. Sakura's classmate, Ino Yamanaka, saw this and defended her from her bullies, encouraging her to embrace her forehead instead of hiding it.
