1、了解多肽的相关信息
1. Understand the relevant information of peptides
通过对多肽序列的分析,参考类似序列的纯化经验,或者查询文献获取有用信息,还可通过了解其酸碱性、亲水性、稳定性等基本性质,来确定纯化过程中的注意事项。
By analyzing peptide sequences, referring to purification experience of similar sequences, or consulting literature to obtain useful information, and understanding their basic properties such as acidity, hydrophilicity, and stability, precautions during the purification process can be determined.
2、相应的监测分析
2. Corresponding monitoring and analysis
通过对多肽粗品进行MS和HPLC检测,我们可以对粗品的情况进行初步的判断,两者相结合大致判断出目标物的出峰位置,进而制定出纯化方法。

3、挑选合适的制备柱

4、多肽粗品的前处理

找到既可使粗品溶解清澈又不会对目标物造成破坏和损伤制备柱的溶剂,然后经过孔径为0.45um的滤膜过滤,得到待上机的清液。
Find a solvent that can dissolve the crude product clearly without causing damage or destruction to the target substance in the preparation column, and then filter it through a filter membrane with a pore size of 0.45um to obtain the clear liquid to be loaded onto the machine.
5、制备方法的确定

6、产物干粉化
