in this video we test for Du
Cells with Du antigen are sensitized with anti-D. This results in the adsorption of anti-D on the surface of the cells without producing hemagglutination (sensitization). The presence of reacted antibody on the surface of Du cells is recognize by using anti human globulin (AHG) which reacts with the coated anybody and brings about hemagglutination, even after anti-D treatment and AHG reaction, the cells are truly Rh-negative.
Requirements :
Glass Test tube
Glass slide
5% cell suspension
Anti-D
Anti human globulin ( AHG)
Incubator or water-bath
Microscope
