Norepinephrine is synthesized when tyrosine comes into the cytoplasm of the neuron along with sodium ions and convert into DOPA by the action of enzyme called as tyrosine hydroxylase and is inhibited by Metyrosine. DOPA is then converted into dopamine by the action of enzyme called as aromatic l-amino acid decarboxylase. Dopamine is transported into the vesicles by VMAT which is driven through the efflux of proton, it is an antiporter. and is inhibited by Reserpine. In vesicle, dopamine is converted into Norepinephrine by action of an enzyme called as dopamine beta hydroxylase. Action potential cause the influx of calcium ions which causes the diffusion of synaptic vesicles into the presynaptic membrane. Adenoceptors, autoreceptors and NET are present for the released NE to act. COMT and MAO degrade NE in synaptic cleft and inside neuron respectively. NET is inhibited by cocaine, tricyclic antidepressents.
