This is lightly based off a story me and a friend are working on. Basically, Bruce knew the man the Joker used to be. They were friends as teenagers and could have possibly been more before Jack/Joker disappeared. Bruce recognizes Jack as the Joker when he publicly demands that Batman demask himself. After Harvey stands up, declaring himself as Batman before Bruce can, Bruce must deal with the Joker while coming to terms with the fact that the man he cared for is gone.
