Internalization of cell surface receptors is followed by their transport to endosomes. A subpopulation of endosomes defined by the presence of the protein APPL, a Rab 5 effector, has a specialized role in propagating signaling of internalized receptors. Here, macropinosomes (arrowheads) generated from membrane ruffles and marked by PI3P (green) are shown to subsequently shed PI3P, and this loss is closely followed by recruitment of APPL1 (red). The association of APPL1 with macropinosomes is transient and terminated as they moved further toward the cell interior.
