One well-known psychoplastogen is ketamine, which has shown promising effects in regrowing dendritic spines and restoring synaptic connectivity. However, ketamine lacks circuit-level selectivity. Professor Olson's lab investigates the serotonin receptor 5-HT2A as a target for modulating neuronal structure. Classic serotonergic psychedelics like LSD and psilocybin have been found to promote dendritic arbor complexity and spine growth in cortical neurons. The effects of psychedelics on neuronal structure are long-lasting, with even short stimulation periods producing sustained changes. Studies have shown that psychedelics like psilocybin can increase dendritic spine density for at least a month. These compounds have shown potential for treating various neuropsychiatric disorders and are believed to target the prefrontal cortex, a critical brain region.
