For a Merchant Request in the Resident Evil 4 Remake (RE4), you need to "Destroy the Blue Medallions". There are a total of 5 Blue Medallions to destroy. Doing so will reward you with Spinel x3 from the Merchant.
This guide shows all the locations of the Blue Medallions in the Farm area, which you'll first come across in Chapter 1.
