Necroptosis and Pyroptosis

Necroptosis and Pyroptosis are two forms of programmed cell death that differ from the traditional apoptosis pathway, playing significant roles in the body's defense mechanisms against infections and in inflammatory responses. Necroptosis is a type of cell death that mimics necrosis but is regulated by specific signaling pathways. Triggered by factors like TNF-alpha, it involves the formation of a complex including RIPK1, RIPK3, and MLKL proteins, leading to cell membrane rupture and release of cell contents, often triggering inflammation. Pyroptosis, on the other hand, is an inflammatory form of programmed cell death primarily triggered by infections. It is characterized by the activation of caspase-1 or caspase-11 in mice (caspase-4 and caspase-5 in humans), which leads to the cleavage of gasdermin D, forming pores in the cell membrane and causing cell lysis. Pyroptosis plays a crucial role in the immune response by eliminating infected cells and releasing inflammatory signals to recruit immune cells.
