This video describes the discovery of PRN1008, an inhibitor of Bruton's tyrosine kinase (BTK), a kinase targeted for oncology, immunology, and inflammation indications. The content of this video was reported by scientists at Principia Biopharma, a subsidiary of Sanofi, in South San Franciso, CA as a Drug Annotations article in the Journal of Medicinal Chemistry. The discovery started with ibrutinib, a known BTK inhibitor, and modified the tether between the scaffold of the drug followed by the functionality of the Michael acceptor. PRN1008 incorporated a distinctive cyanoacrylamide that allowed reversible, covalent inhibition of BTK. PRN1008 has advanced into clinical trials for the treatment of both pemphigus vulgaris and immune thrombocytopenia.
