In this short guide, we introduce rabies virus and discuss how this highly specific and efficient virus infects the host nervous system. We then discuss the changes we can make to the virus to perform synaptic tracing: by adding EGFP and removing the viral G glycoprotein from the viral genome, we can perform monosynaptic tracing when the G protein is provided in trans. We can further modify the viral capsid using the TVA/EnvA system to more precisely control initial infection with the recombinant virus.
