Enjoy a brief history of the discovery, and then development, of antidepressant and anxiolytic compounds, the first-generation TCAs and MAOIs that ushered in the era of psychopharmacology, and the second-generation agents, which heralded the start of the modern age of specific, selective psychotropic medications, beginning with Prozac in 1987 (in the United States).
But, if the tricyclic agents belong to the first generation, and the SSRIs and modern SNRIs belong to the second, where do tetracyclic compounds like mianserin, maprotiline, mirtazapine and amoxapine fit in? Some classification schemes lump them with the first generation agents, despite their novel chemical structures and mechanisms of action, while others consider them "second-generation," though they bear no resemblance to Prozac, et al., either.
This video is a deep-dive into the unique mechanisms of action of mianserin and it's analogue mirtazapine (the latter of which is also always referred to as a "second-generation antidepressant"!).
